Özet

This study presents an integrated pharmacokinetic-pharmacodynamic (PK-PD) modeling framework for multi-drug chemotherapy used in pediatric Acute Lymphoblastic Leukemia (ALL). The model addresses the pharmacological effects of three commonly used drugs (6-mercaptopurine (6-MP), methotrexate (MTX), and vincristine (VCR)) simultaneously within a unified mathematical structure. For each drug, PK models describing absorption, distribution, metabolism, and elimination processes are combined under a drug effect function and integrated into the overall system. The model predicts hematological responses based on white blood cell (WBC) and absolute neutrophil count (ANC), as well as vincristine-induced peripheral neuropathy (VIPN) indicators. The simulation results demonstrate treatment response by capturing interactions between drugs and patient-specific variables across a four-phase treatment protocol, and provide a computational basis for personalized dose optimization in pediatric ALL treatment.
