Determination of the dissociation constants of six pharmacologically active imidazole antifungal agents by RPLC method

In the present study, the dissociation constant () of hydroorganic mixtures of climbazole, lanoconazole, bifonazole, butoconazole, oxiconazole, and sertaconazole, which have low water solubility containing imidazole groups, was determined for the first time using the developed reversed-phase liquid chromatography (RPLC) method. Analyses were carried out on binary mixtures of water and acetonitrile containing 35–55 % (v/v) acetonitrile on a Pinnacle DB Cyano (250 × 4.60 mm, 5.0 μm) column at 25 °C. The experimental retention data were modeled to match the theoretical retention data using the solvatochromic parameter (). The pKa () values of these low water solubility compounds were calculated using the mole fraction and dielectric constant values of acetonitrile used in the mobile phase. Using these data, the ionization percentages of the compounds were calculated using the Henderson-Hasselbalch equation. Additionally, the values of these antifungal agents at 37 °C were estimated according to the Abraham solvation parameter model.
