Origanum sect. Amaracus Türlerinde Kemotipik Benzerlikler Filogenetik İlişkileri Yansıtıyor mu?

Origanum
L. sect. Amaracus (Gled.) Vogel, Doğu Akdeniz’de yayılış ve çeşitlenme
gösteren, sekiz tür (O. ayliniae Dirmenci
& T.Yazici, O. boissieri Ietsw., O. calcaratum Juss., O. cordifolium (Montbret & Aucher ex Benth.) Vogel, O. dictamnus L., O. saccatum P.H.Davis, O. solymicum P.H.Davis, O. symes Carlström) içeren önemli bir
seksiyondur [1-2]. Türler arasındaki filogenetik ilişkiler ile uçucu yağ
bileşimine dayalı kemotipik farklılıkların birlikte değerlendirilmesi, bir
cinsin sistematik ilişkilerinin ve fitokimyasal çeşitliliğinin daha kapsamlı
biçimde ortaya konulmasına katkı sağlamaktadır [3]. Bu çalışmada, Origanum
sect. Amaracus türlerinde uçucu yağ bileşimi ile ITS bölgesine dayalı
filogenetik ilişkiler arasındaki uyumun değerlendirilmesi amaçlanmıştır. Literatürden
derlenen uçucu yağ bileşimi verileri temel alınarak Principal Component
Analysis (PCA) uygulanmış, filogenetik ilişkiler ise ITS dizilerine dayalı
Neighbor-Joining yöntemi ile değerlendirilmiştir. PCA sonucunda ilk iki bileşen
toplam varyansın %85,15'ini açıklamıştır. PC1 ekseni başlıca p-simen ve
γ-terpinen ile pozitif, karvakrol ile negatif yük göstermiştir. Buna göre O.
saccatum ve O. solymicum p-simen ve γ-terpinen
bakımından benzer kemotipik özellikler sergilerken, O. dictamnus, O.
ayliniae ve O. boissieri karvakrol ağırlıklı profilleriyle
ayrılmıştır. PC2 ekseni ise büyük ölçüde timol tarafından temsil edilmiş ve
yüksek timol içeriği nedeniyle O. calcaratum diğer türlerden
farklılaşmıştır. Neighbor-Joining analizinde O. ayliniae–O. saccatum
(BS=95), O. boissieri–O. symes (BS=75) ve O. dictamnus–O.
solymicum (BS=64) arasında yakın filogenetik ilişki belirlenmiştir. PCA
sonuçları ile ITS temelli filogenetik ilişkiler arasında tam bir uyum
gözlenmemiştir. Elde edilen bulgular, Origanum sect. Amaracus
türlerinde kemotipik benzerlikler ile filogenetik ilişkiler arasında tam bir
uyum bulunmadığını göstermektedir. Bu durum, uçucu yağ bileşiminin
değerlendirilmesinde filogenetik ilişkilerin tek başına yeterli
olmayabileceğine işaret etmektedir.
Origanum
L. sect. Amaracus (Gled.) Vogel is an important section distributed and
diversified in the Eastern Mediterranean, comprising eight species (O.
ayliniae Dirmenci & T. Yazici, O. boissieri Ietsw., O.
calcaratum Juss., O. cordifolium (Montbret & Aucher ex Benth.)
Vogel, O. dictamnus L., O. saccatum P.H. Davis, O. solymicum
P.H. Davis, and O. symes Carlström) [1–2]. The combined evaluation of
phylogenetic relationships among species and chemotypic differences based on
essential oil composition contributes to a more comprehensive understanding of
the systematic relationships and phytochemical diversity of a genus [3]. In
this study, it was aimed to evaluate the concordance between essential oil composition
and ITS region-based phylogenetic relationships in Origanum sect. Amaracus.
Principal Component Analysis (PCA) was performed based on essential oil
composition data compiled from the literature, while phylogenetic relationships
were evaluated using the Neighbor-Joining method based on ITS sequences. The
first two principal components explained 85.15% of the total variance. The PC1
axis showed positive loadings mainly for p-cymene and γ-terpinene, and a
negative loading for carvacrol. Accordingly, O. saccatum and O.
solymicum exhibited similar chemotypic characteristics in terms of p-cymene
and γ-terpinene, whereas O. dictamnus, O. ayliniae, and O.
boissieri were distinguished by their carvacrol-dominant profiles. The PC2
axis was largely represented by thymol, and O. calcaratum was
differentiated from the other species due to its high thymol content.
Neighbor-Joining analysis revealed close phylogenetic relationships between O.
ayliniae–O. saccatum (BS = 95), O. boissieri–O. symes
(BS = 75), and O. dictamnus–O. solymicum (BS = 64). No complete
concordance was observed between the PCA results and the ITS-based phylogenetic
relationships. The findings indicate that there is no complete concordance
between chemotypic similarities and phylogenetic relationships in Origanum
sect. Amaracus. This suggests that phylogenetic relationships alone may
not be sufficient for the evaluation of essential oil composition.
