Suppression of Lung Cancer Cell Motility and Growth by Lycopene via MMP-2/9 Modulation

Özet

Lycopene, a naturally occurring carotenoid, has attracted attention for its potential anticancer properties. In this study, t he anti-proliferative and anti-migratory effects of lycopene were evaluated in A549 human lung adenocarcinoma cells. Cell viability was assessed using the MTT assay following 48-hour exposure to increasing concentrations of lycopene (1.96875– 63 µg). Lycopene exhibited a dose-dependent reduction in cell viability, with an IC₅₀ value of 22.57 μM. The effect of lycopene on cell migration was investigated by a wound healing assay. Treatment with 22.57 μM lycopene significantly inhibited cell migration compared to the control group (P < 0.001), with a time-dependent increase in i nhibitory effect. To further explore the underlying mechanisms, the levels of matrix metalloproteinases MMP-2 and MMP-9 were quantified by ELISA. Lycopene treatment significantly reduced MMP-2 and MMP-9 levels by approximately 20% and 24%, respectively (P < 0.001). These findings suggest that lycopene suppresses both proliferation and migration of A549 cells, potentially through downregulation of MMP-2 and MMP-9, highlighting its potential as a therapeutic agent in lung cancer.
