YÜKSEK FRUKTOZLU MISIR ŞURUBUNA BAĞLI GELİŞEN KARDİYOVASKÜLER HASARDA SELENYUM’UN İYİLEŞTİRİCİ ETKİSİ

Özet
Amaç: Günümüzde
yaygın olarak hazır gıdalarda kullanılan yüksek dozlu fruktoz şurubunun (YFMŞ)
oksidatif stres ve inflamasyona bağlı olarak kardiyovasküler sistem üzerine
toksik etkilerinin olduğu bildirilmiştir. Selenyum (SEL), anti-inflamatuar ve
antioksidan özellik gösteren bir eser elementtir. Bu çalışmada YFMŞ’nin oluşturduğu
kardiyovasküler toksisitede SEL’in olası koruyucu veya iyileştirici etkilerini
araştırmak hedeflenmiştir.
Gereç-Yöntem:
Kırk adet Wistar albino cinsi dişi sıçan rastgele 4 gruba
ayrıldı. 1) Kontrol grubu: Rutin beslenme programı uygulanmış olup günlük
sularına hiçbir etken madde eklenmedi. 2) YFMŞ grubu: %30’luk fruktoz çözeltisi
içme suyuna %30 oranında karıştırıldı. 3)YFMŞ + SEL grubu: %30’lik fruktoz
çözeltisi içme suyuna %30 oranında karıştırıldıktan sonra 0,3 mg/kg SEL içme
suyuna ilave edildi. 4) SEL grubu: 0,3 mg/kg SEL içme suyuna ilave edildi. 6
haftanın sonunda sıçanlar sakrifiye edilerek kalp ve endotel dokuları alındı. Dokular
rutin Hematoksilen Eozin boyama ile değerlendirildi. İmmünohistokimyasal
analizle kaspaz-3 (Cas-3), tümör nekroz faktör-alfa (TNF-α), interlökin-1β
(IL-1β) ve vasküler endotelyal büyüme faktörü (VEGF) belirteçleri değerlendirildi.
Bulgular:
Histopatolojik analizlerde YFMŞ grubuna ait kalp dokularında
hiperemi ve ödem gözlendi. Buna paralel olarak endotel hücrelerinde kayıp
saptandı. SEL grubunda bu patolojilerde düzelme elde edildi. İmmünohistokimyasal
analizler sonucunda YFMŞ grubunda ekspresyonları artan Cas-3, TNF- α, IL-1β ve
VEGF düzeyleri SEL tedavisi ile anlamlı şekilde azaldığı gözlemlendi (p<0.001).
Sonuç: YFMŞ
ile oluşturulan sistemik inflamasyon modelinde meydana gelen kalp ve endotel dokularında
oluşan patolojik bulgular, anti-inflamatuar etkinliği olan SEL ile iyileşme
gösterdi. Bu nedenle çalışmamızda elde ettiğimiz veriler doğrultusunda SEL’in
YMFŞ’ye bağlı kardiyovasküler hasar ve buna benzer toksisite durumlarında
kullanılabilecek güçlü bir aday olabileceği düşünülmektedir.

Objective: It has
been reported that high-dose fructose syrup (HFCS), which is widely used in
convenience foods, has toxic effects on
the cardiovascular system via oxidative stress and inflammation. Selenium (SEL)
is a trace element with anti-inflammatory and antioxidant properties. In this
study, we aimed to investigate the possible protective or ameliorative effects
of SEL in cardiovascular toxicity caused by HFCS.
Materials-Methods: Forty
Wistar albino female rats were randomly divided into 4 groups. 1) Control
group: Routine nutrition program was applied and no active substance was added.
2) HFCS group: 30% fructose solution was mixed into drinking water at a rate of
30%. 3) HFCS+SEL group: After mixing 30% fructose solution with 30% drinking
water, 0.3 mg/kg SEL was added to drinking water. 4) SEL group: 0.3 mg/kg SEL
was added to drinking water. At the end of 6 weeks, rats were sacrificed and
their heart and endothelial tissues were collected. Samples were evaluated by
routine Hematoxylin-Eosin staining. Caspase-3 (Cas-3), tumor necrosis
factor-alpha (TNF-α), interleukin-1β (IL-1β) and vascular endothelial growth
factor (VEGF) markers were evaluated immunohistochemically.
Results:
Histopathological analyzes revealed hyperemia and edema in the heart tissues of
the HFCS group. Parallel to this, loss of endothelial cells was detected. In
the SEL group, these pathologies improved. As a result of immunohistochemical
analysis, it was observed that Cas-3, TNF-α, IL-1β and VEGF levels, whose
expressions were increased in the HFCS group, decreased significantly with SEL
treatment (p<0.001).
Conclusion:
Pathological findings in the heart and endothelial tissues in the systemic
inflammation model created by HFCS improved with SEL, which has anti-inflammatory
activity. Therefore, in line with the data we obtained in our study, we thought
that SEL may be a strong candidate to be used in cases of cardiovascular damage
and similar toxicity due to HFCS.
